Hm, it depends on how large each tile is actually meant to be. In my imagination, a tile is more than large enough to have room for any city (and huge amounts of soldiers, so I am not convinced by the one-unit-per-tile of Civ V).

Hmm, but a lot of cities have suburbs that provide crucial supplements towards the city itself, essentially becoming part of the cities eco-system.
